Bioanalytical techniques are a major focus of the "Protein Synthesis & Bioanalytics" core facility. A range of techniques that span from general bioanalytical applications to more specialized requirements are offered and undertaken.

LC-20A Recycling HPLC System (Shimadzu)

For very complicated compounds the innovative recycling function of our LC-20A HPLC system can be used to produce results at higher resolutions.

LC-MS/MS

LC-MS/MS can be used to confirm or analyze the molecular weight as well as the composition of various compounds. Subsequent application of ESI-MS/MS to tryptic digest is another standard method offered by the core facility. The core facility uses the LC-ESI micrOTOF-Q III mass spectrometer (Bruker Daltonics GmbH).

Amino acid analysis

Amino acid analysis is regularly used to determine peptide and protein concentrations and composition. This is one of the most precise techniques that is still applied in food chemistry. The core facility uses an Eppendorf-Biotronik LC 3000 system (Laborservice Onken).

Edman degradation (N-terminal protein sequencing)

Edman degradation is used in bioanalytics for sequencing of peptides and proteins by stepwise cleavage of the N-terminal amino acids. Furthermore, post-translational modifications of these amino acids can be identified by this sequencing method. Therefore, the core facility uses a PPSQ-53A protein sequencer (Shimadzu).

Additional techniques

Additional chromatographic and spectroscopic techniques can be requested to complete the analytical profile of the compounds of interest. Some of the techniques offered in this regard are listed below

  1. Thin layer chromatography

  2. UV/Vis-Spectroscopy (Cuvette or Microplate reader)
